Resumo
The biosynthesis of PGF2a and PGE2 can be altered by specific fatty acids, including conjugated linoleic acid (CLA). Decreased PGF2a synthesis and/or increased PGE2 synthesis and PGE2/PGF2a ratio may favor maternal fetal recognition in female beef cattle. The hypothesis is that supplementation with CLA (Sigma-Aldrich, USA, Cat no. O5507) during in vitro embryo culture (IVC) promotes, in the 9-day-old embryo, a decrease in the synthesis of PGF2a and an increase in the synthesis of PGE2 and in the PGE2/PGF2a ratio, such effect being dose dependent. The objective was to determine the effects of different concentrations of CLA (0, 50, 100 and 150 µM) added to the IVC medium, on the concentrations of PGF2a and PGE2 and the PGE2/PGF2a ratio in the culture medium at the end of the IVC. Oocytes obtained by follicular aspiration of ovaries from slaughtered Nellore cows were selected to compose one of the four treatment groups, where the IVC medium contained: absence of CLA (Control Group; n = 131); 50 µM CLA (CLA Group 50 µM; n = 131); 100 µM CLA (CLA Group 100 µM; n = 132) or 150 µM CLA (CLA Group 150 µM; n = 130). From 12 to 28 oocytes were used per treatment and per experimental repetition, totaling 8 repetitions. The cultivation of oocytes and embryos was carried out in 4-well plates containing 500 µL of the medium corresponding to each treatment. After 9 days of IVC (D9; D0 = day of in vitro fertilization) the culture medium was recovered, frozen in microtubes and subsequently analyzed by ELISA. Statistical analyzes were performed using PROC MIXED from SAS considering the well as an experimental unit. At the end of the IVC, PGF2a concentrations were lower (P = 0.0005) in the 50 µM (0.1624 ± 0.0425 ng/mL), 100 µM (0.1220 ± 0.0442 ng/mL) and 150 µM (0.0988 ± 0.0442 ng/mL) compared to the Control Group (0.4163 ± 0.0442 ng/mL). PGE2 concentrations were lower (P = 0.0005) in the 50 µM (0.5025 ± 0.2197 ng/mL), 100 µM (0.3273 ± 0.2197 ng/mL) and 150 µM groups (0.2740 ± 0.2197 ng/mL) compared to the Control Group (1.4799 ± 0.2197 ng/mL). The PGE2/PGF2a ratio did not differ between groups (P = 0.9087). A higher rate of blastocysts was observed on D9 in the group treated with 50 µM compared to 150 µM (40.8978 ± 7.003% vs. 8.6411 ± 7.003%; P = 0.4081), although both did not differ from Control Group. It is concluded that supplementation with CLA during IVC promotes a decrease in the synthesis of PGF2a and PGE2 in the embryo, without altering the PGE2/PGF2a ratio.
